What is Fulvestrant?
Fulvestrant is a drug used to treat hormone receptor (HR)-positive metastatic breast cancer and HR-positive and HER2-negative advanced breast cancer in postmenopausal women whose disease has progressed. It is used in combination with palbociclib in women whose disease has progressed after endocrine therapy. . It is injected into the muscle.
Fulvestrant is a selective estrogen receptor degradant (SERD), the first of its kind. Its role is to bind to the estrogen receptor and make it unstable, causing the cell’s normal protein degradation process to destroy it.
Fulvestrant was approved for medical use in the United States in 2002.
Medical use
Fulvestrant is a valuable option with limited toxicity and long-lasting response in metastatic HER2 and ER-positive breast cancer that has progressed with anti-HER2 drugs. Even in extensive skin metastases and (irradiated) brain metastases, therapeutic benefits are expected.
Side effects
Very common (occurring in more than 10% of people) adverse reactions include nausea, injection site reactions, weakness, and elevated transaminases. Common (1% to 10%) adverse reactions include urinary tract infections, allergic reactions, loss of appetite, headaches, venous thrombosis, hot flashes, vomiting, diarrhea, elevated bilirubin, skin rashes, and back pain. In a large clinical trial, the incidence of venous thromboembolism (VTE) of Fulvestrant was 0.9%.
