What is Medroxyprogesterone?
Medroxyprogesterone, also known as the injectable form of long-acting medroxyprogesterone acetate (DMPA), is a progesterone hormone drug. It is used as a contraceptive method and part of menopausal hormone therapy. It is also used to treat endometriosis, abnormal uterine bleeding, abnormal male sexual behavior, and certain types of cancer. The drug can be used alone or in combination with an estrogen.
Medroxyprogesterone was discovered in 1956 and introduced for medical use in the United States in 1959. It is on the list of essential medicines of the World Health Organization. MPA is the most widely used progestin in menopausal hormone therapy and birth control using only progesterone. DMPA is approved as a long-acting contraceptive method in more than 100 countries. In 2018, it was the 202nd most commonly used prescription drug in the United States, with more than 2 million prescriptions.

When combined with CEE, Medroxyprogesterone increases the risk of breast cancer, dementia, and eye thrombosis. Usually used in combination with estrogen, Medroxyprogesterone may increase the risk of cardiovascular disease, and the correlation is stronger when postmenopausal women also take CEE. It is precise because of these unexpected interactions that the Women’s Health Initiative study ended early due to the additional risks of menopausal hormone therapy, resulting in a sharp decrease in new and updated prescriptions for hormone therapy.
Long-term studies of DMPA users have found that the overall risk of breast cancer is slightly increased or not. However, the study population does show that recent users under the age of 35 (who used DMPA in the past four years) have a slightly increased risk of breast cancer, similar to those seen when using combined oral contraceptives.
