What is Oleamide (ODA)?
Oleamide is an organic compound, which is an amide derived from fatty acid oleic acid. It is a colorless waxy solid that exists in nature. Sometimes labeled as a fatty acid primary amide (FAPA), it is biosynthesized from N-oleoylglycine.
Medical use
Biochemical and medical aspects
In terms of natural occurrence, oleamide was first detected in human plasma. It was later shown to accumulate in the cerebrospinal fluid during sleep deprivation and induce sleep in animals.
It is considered a potential treatment for mood and sleep disorders and cannabinoid-regulated depression.
In terms of its sleep-inducing effect, it is speculated that oleamide interacts with a variety of neurotransmitter systems.
Withdrawal
The potentially addictive properties of oleamide have been studied because it is known to enhance GABAA receptor signaling, which is the pharmacological effect of benzodiazepines (through the benzodiazepine binding site located on the GABAA receptor.
A trial in which rats were given oleamide for 10 days (30 mg/kg twice a day) and then stopped treatment suddenly failed to find evidence of withdrawal because there was no difference in withdrawal symptoms between the oleamide condition and the control group. Co-administration of CB1 antagonists with the above-mentioned dosing regimen seems to cause withdrawal symptoms, but their levels are lower than opioids and anandamide.
Compared with control rats, sudden cessation of oleamide after 15 days does not seem to have a negative effect on sleep, although the enhancement of REM sleep observed with oleamide treatment disappeared.
